Abstract

Sertralinedrug (STH)is currently one of the mostpopular antidepressants in Europe and in the United States, ranking 11th of all drugs prescribed in the US. It is found as hydrochloride.It is considered a member of selective serotonin reuptake inhibitors (SSRIs) family. Although, SSRIs are considered the second generation of antidepressant drugs, they are becoming the drugs of first choice for the treatment of depression. The successful introduction of SSRIs was indeed a milestone in the history of pharmacotherapy for depression.
Sertraline antidepressant drug is also used in the treatment of anxiety related disorders (obsessive compulsive disorder, social anxiety, and panic disorders), autism, eating disorders, occasionally posttraumatic stress disorder and menopausal symptoms. SSRIs all act similarly by blocking serotonin transporters, thereby preventing the reuptake of serotonin (5-hydroxytryptamine, 5-HT)from the synaptic cleft into the presynaptic serotonergic neuron. This leads to elevated extracellular 5-HT levels. In rodents, the blockade of the 5-HTtransporter alters emotional behavior in adult mice.
Serotonin is an essential neurotransmitter for normal function in both the central and peripheral nervous systems. Increased synaptic availability of serotonin stimulates a large number of postsynaptic 5-HT receptor subtypes which lead to complex secondary responses including gastrointestinal disturbances, nausea, vomiting and sexual side effects which include loss or decrease of libido, delayed ejaculation, anorgasmia and impaired orgasm, decreased testicular development and decreased Sertoli cells populations, which may lead to infertility in adults.The present study attempts to evaluate the effect of oral treatment with sertraline drug on the fertility parameters of male Wister rats.
One hundredand sixty new weanling male Wister rats (21 days old) were acclimatized to the laboratory environment for two weeks prior to the start of experiments. The experiments started when rats reach to 35 days old and weighing 55-57 g. After the period of acclimation,animals were randomly divided into four groups (GI, GII, GIII, and GIV), each group subdivided into a control and a treated subgroup (SGI, SGII, SGIII, and SGIV).
