Abstract

The present study deals with the development of the physical gene map of river buffalo by determining the synteny of some genes and postulating their assignment to specific chromosomes.
Twenty-nine independent buffalo - hamster hybrids, obtained by the fusion of blood lymphocytes from Egyptian water buffaloes with a HPRT- Chinese hamster cell line (wg 3h cl2), were used to
examine synteny between studied genes, which code for the
following isozymes; MEl, PGM3, NP, PKM2, ADA and ITPA.
Starch gel or cellulose acetate electrophoresis was used to determine the presence or absence of gene products in the different hybrids and their segregations were recorded. Synteny among the six genes was investigated by calculating the percent concordance and correlation coefficient (cp). The analysis revealed the presence of the following syntenic groups: ME1-PGM3 and NP-PKM2. The synteny between ADA and ITPA was also speculated.
MEl and PGM3 cosegregate with a 93.1% concordance and cp = 0.83. MEl and PGM3 were found to segregate independently from the other four genes as the highest cp value did not exceed 0.31, which was recorded with NP.
The genes coding for NP and PKM2 were found to cosegregate with 92.6% concordance and cp = 0.85. They were also found to segregate independently from the other genes under investigation. The highest cp value recorded was only 0.31 with
MEl.
