Abstract

This study was conducted on 5 sheep flocks (1450 animals) located at Alexandria and Marsa Matrouh Governorate. Animals were reared in mobile flocks to determine Epidemiological situation, Clinical observation, Molecular diagnosis and Antibiotics resistence of Corynebacterium pseudotuberculosis.
Clinical observation of caseous lymphadenitis in 24 sheep characterized clinically by emaciation, unthriftness and enlargement of parotid, retropharyngeal, sub mandibular, pre scapular and inguinal lymph node with characteristic greenish pus.
Parotid, retropharyngeal, and sub mandibular lymph node consider the most common lymph node affected by (87.5%, 21 case), followed by pre scapular lymph node (8.33%, 2 case), followed by inguinal lymph node (4.16%, one case).
External abscess was recorded in 42 animals out of 1450 Animals (2.8%).
No of infected sheep with of Corynebacterium pseudotuberculosis was 24 cases (1.65%) (Corynebacterium pseudotuberculosis alone (10 cases) with strept pyogens (4 cases) with Staphylococcus aureus (10 cases). Staphylococcus aurous from 11 cases (0.7%). Strept pyogens from 3 cases (0.20%). Staphylococcus aureus with strept pyogens in 4 cases (0.27).
The infection rates of caseous lymphadenitis were zero percent in One month age till 12th month These results may related of partial protection by maternal antibodies or because they have not had the same length of exposure to bacteria or a factor of long incubation period. While 1.80% (4 animals), 1.18% (3 animals) 4.54% (8 animals), 9.1% (9 animals), in age group 1-2 years old (222 animals), 2-3 years old (254 animals), 3-4 years old (176 animals) and 4-5 years old (98 animals), respectively.
The infection rates of caseous lymphadenitis were more prevalent in femals 2.24% (22 animals) than males 0.42% (2 animals).
We concluded that molecular diagnosis (PCR) in cultured samples has proved to be more efficient, reproducible and faster than PCR on pus to overcome the obstacles of pus and necrotic tissue.
PCR confirm C. pseudotuberculosis diagnosis, and the best contribution for the epidemiological surveillance of the disease in sheep.
Corynebacterium pseudo tuberculosis more sensitive to, Marbofloxacine, Florofenicole and Enrofloxacin. Sensitivity of Corynebacterium pseudotuberculosis to antibiotics not indicative to control the disease due to chronic infection, internal lesion and presence of fibrous capsule which prevent contact between the drug used and the pathogen inside the host.
