Abstract

Topical application of ophthalmic drugs is the method of choice for treatment of ophthalmic disorders because it is convenient and safe. Among these systems, niosomes were selected to design sustained ocular delivery systems for flurbiprofen. Niosomes are a versatile drug delivery systems that used to solve some major biopharmaceutical problems including bioavailability. It was found that flurbiprofen niosomes prepared using span 60 showed the highest entrapment efficiency than most of other surfactant used. EE% of the prepared niosomes increased with increasing drug concentration, and total lipid content. Then evaluation of an ophthalmic delivery system of flurbiprofen, based on the incorporation of niosomes in carbopol gel system have been done. Niosomes were incorporated in carbopol gel in order to increase the precorneal residence time of a niosome suspension. Also evaluation of the produced niosomes and niosomes incorporated in carbopol gel in an animal model rabbits have been done. The results showed that Ocufen® achieved the therapeutic level of flurbiprofen only during the first two hours after instillation. While the FBP niosomes loaded carbopol gel showed the longest duration of action during the first 12 hs after instillation.
