Abstract

Foot and mouth disease virus is severe, highly contagious pathogen of cloven-hoofed animals that affects large animal livestock species and molecular characterization of FMDV is essential for effective control strategies of the disease produced by this virus In this study a total number of 35 field samples ( 31 tongue epithelial tissues and 4vesicular fluid samples) were collected from clinically diseased cattle and buffaloes in different governorates in north of Egypt (Alexandria, Behera and Kafrel-sheikh) during the outbreak from April to September 2015 SerotyPing of collected samples using Antigen-detection ELISA revealing that 26(74.23%) of them were positive for serotype O RNA extracted from representative samples from them and performing real-time RT-PCR protocol on them using universal probe for FMDV detection revealing a positive results Serotyping of theses samples by RT-PCR using serotype-specific primers revealing positive results for serotypes (O ) Sequencing of the VP1-coding region followed by phylogenetic tree of the serotyped samples revealing that serotype O is closely related to O Sharquia/EGY/2014, O /3/ GiZa/ EGY/2014, O /2/ GiZa/ EGY/2014, O /Fayoum/ EGY/2014 and O /Qaliubia/ EGY/2013 of East Africa-3 (EA-3) topotype which not detected in Egypt before so it should be included in the vaccines locally produced in Egypt for complete protection against FMDV infection.
