Abstract 50 sputum samples were collected from patients suspected of pulmonary tuberculosis through X-ray radiography and Mantoux test over a period of 9 months. The demographical data of the patients was recorded and analyzed. 50 sputum samples were smear positive for acid fast bacilli. Out of the 50 smear positive samples, 24 samples only gave culture positive on LJ media. It was found that the prevalence of TB was higher in female and was more common in patients aged 21-40 years .our results revealed that smoking, family history, presence of underlying diseases (Diabetes mellitus, hypertension) were important factors for the development of tuberculosis disease .Also prevalence of pulmonary tuberculosis infection was the highest among housewives especially in rural area. All grown isolates were identified as M .tuberculosis according to physical characters, rate of growth, biochemical tests. All isolated strains gave nearly the same amplicon size when amplify350 bp region of plc by PCR. Direct sequencing analysis of the PCR amplified PLC gene revealed many substitution mutations along the entire sequenced fragment. The phylogenetic tree subcladed the seven sequenced isolates into three distinct nodes. |
