Abstract This work was undertaken to study the histological, histochemical and the ultrastructural changes that might occurred in the renal cortex of adult male albino rats after injection of large single dose of amikacin and also to study the possible effect of montelukast on those changes. Fifty adult male albino rats with average weight of (150-200gm) were used in this study and divided into five equal groups. Group I: control group (animals were given 2cm milk by gastric tube once a day for 10 days). Group II: the animals were given montelukast (10 mg/kg) dissolved in 2cm milk by gastric tube once a day for 10 days. Group III: the animals were injected, intraperitonelly, by a single dose of amikacin (1.2 g/kg). Group IV : the animals were injected, intraperitonelly, by amikacin (1.2 g/kg) like group III and after 3 days of amikacin injection, they were given montelukast (10 mg/kg) dissolved in 2cm milk by gastric tube once a day for 10 days. Group V : the animals were given montelukast (10 mg/kg) dissolved in 2cm of milk by gastric tube once a day for 10 days. After the last dose of montelukast, they were injected, intraperitonelly, by a single dose of amikacin (1.2 g/kg) Three days after the last dose in all groups the animal was weighed and both kidneys were dissected and weighed. Also blood samples were obtained from all animals to determine serum creatinine levels. Kidney samples from rats of each group were taken to prepare paraffin sections to be stained with H&E and PAS reaction. Also fresh frozen sections were prepared and chemically processed for localization of alkaline phosphatase activity in the renal tissue. Furthermore, small specimens from the kidney were taken and prepared to get semi-thin sections stained by toludine blue and other ultrathin sections for examination by transmission electron microscope. The animals in group IV appeared response to the drug while group V shown no response. |
