The present work was carried out to study the biological activity of aqueous extract
and shoot crude powder of Deverra tortuosa (DTSAE and DTSCP) and Haplophyllum
tuberculatum (HTSAE and HTSCP), as well as a mixture (w/w) of both on some
germination and growth parameters (germination bioassay experiment) besides major
physiological, biochemical and molecular processes (pot experiment) in Triticum aestivum
(crop species) and Phalaris minor (weed species) in pure and mixed cultures.
Generally, the effect of the extracts in pure and mixed cultures on germination
percentage (GP), seed germination index (SGI), energy of germination (GE), coleoptile
(CL) and radicle (RL) lengths was arranged in the following order: HTSAE > DTSAEHTSAE
mixture > DTSAE. In pure culture, GP of T. aestivum seeds was ineffectively
decreased with increasing the concentrations of DTSAE and HTSAE while the percentage
was significantly decreased with increasing the concentrations of the two types of the
extracts in mixed culture with P. minor. SGI and GE of T. aestivum seeds in pure culture
decreased as concentration of DTSAE, HTSAE and their mixture increased. The same
results with notable decreasing were obtained in mixed culture with P. minor. All
concentrations levels of the extracts reduced coleoptile (CL) and radicle (RL) lengths of T.
aestivum in pure and mixed cultures. Likewise, the reduction in the two parameters was
documented in case of pure and mixed cultures related to P. minor.
Treatment with HTSCP or the mixture HTSCP+DTSCP had a more negative effect
on soluble protein content in P. minor than on T. aestivum. The allelopathic compounds
released from HTSCP and DTSCP significantly suppressed the uptake of almost all
elements studied. The two recipient plants exhibit a tendency to accumulate amino acids
and free proline. Control T. aestivum seedlings grown in mixed culture had higher total
antioxidant content compared to seedlings grown in pure culture. There was a significant
increase in antioxidants in T. aestivum seedlings treated with 40% DTSCP. The reduction
in total photosynthetic pigment content of the two recipient seedlings upon treatment with
DTSCP, HTSCP or the mixture of both could be attributed to the decrease in both Chl.a
and Chl.b.
The control seedlings of T. aestivum grown in pure culture revealed the presence of
six peptides with molecular masses ranging from 185 to 10 kDa. Treatment with a mixture
of both DTSCP and HTSCP however induced the formation of 11 protein bands in T.
aestivum seedlings. In these seedlings high and low molecular masses proteins of 200, 199,
198, 189, 169, 162, 137, 126, 125, 79 and 1 kDa were synthesized in response to this
treatment. Several peptides (11) with different band intensities were present in P. minor
control seedlings grown in mixed culture. Treatment with DTSCP and HTSCP revealed the
presence of only 4 and 5 protein bands respectively. Application of HTSCP to P. minor
grown in pure culture revealed the presence of six bands. Meanwhile.
