Abstract

Cisplatin is one of the well known chemotherapeutic agents, but it has a high toxicity on normal tissues as well as having nephrotoxicity during cancer therapies. This study aims to investigate the effect of feeding normal and renal failure rats on diets contain one source of protein (either casein (C) or one from tested legumes, namely; fenugreek (F), lupin (L), soybean (S)) supplemented with 5% black seeds (BS) only or with either 5% barley (B) or 5% guava seeds (GS). Rats were injected with cisplatin intraperitoneal (i.p.) at a dose of 3mg/kg b.w to induce a reduction in kidney function as a model of renal failure. The used dose from cisplatin was repeated after 3 weeks from the first dose. 112 male rats of Sprague Dawely strain were randomly classified into treated and protected renal failure groups. The treated group were injected with saline or cisplatin, and then fed on experimental diets to 8 weeks. The protected group fed on the experimental diets for one week, and then injected with cisplatin and continued feeding on these diets to 8 weeks. Both these groups were randomly classified to control (-ve) group which injected with saline (i.p.) and fed on standard diet and renal failure group which were fasted for 12h and then injected with cisplatin. Renal failure group in both treated and protected group was reclassified into 13 groups (4 rats each) as follows: G1Control (+ve), G2 (C+BS), G3 (F+BS), G4 (L+BS), G5 (S+BS), G6 (C+BS+B), G7 (F+BS+B), G8 (L+BS+B), G9 (S+BS+B), G10 (C+BS+GS), G11 (F+BS+GS), G12 (L+BS+GS) and G13 (S+BS+GS). The results revealed that cisplatin administration caused in a significant decrease in both of weight gain, food efficiency ratio, liver and kidney antioxidant enzymes. Also, administration of cisplatin caused in a significant increase in creatinine, urea, uric acid, globulin and liver functions parameters in serum of positive control group when compared with their corresponding values in normal control (-ve) group. In addition, it was found that feeding on the tested diets especially soybean diet which supplemented with (BS) and (GS) was the best among the tested diets in improvement both of the nutritional indicators, blood picture, liver and renal function parameters, liver and renal antioxidant enzymes, serum minerals and pH, and weight and relative weight (%) of organs values in both treated and protected renal failure rats. This may be due to the combine effect of the nutritional and phytochemical components of both of soybean, black and guava seeds which may provide a partial protection against cisplatin-induced structural and functional alterations in rats. According to the results of this study, it is recommended with addition soybean, black and guava seeds in the diet of cancer patient who treated by cisplatin and renal failure patient for improvement their health.
