Abstract Colon cancer (CC) is a world health problem. Aspirin ( a cyclooxygenase inhibitor) has been shown to reduce the risk of CC . The present study was designed to evaluate the effect of aspirin and luteolin (LUT) either alone or combined with low dose gamma irradiation on 1,2 dimethylhydrazine (DMH) in rats. DMH (20 mg/kg BW/ week) injected rats gavaged aspirin (50 mg/ kg BW/ week) asn LUT (0.2 mg/kg Bw/ day) and exposed to (0.5 Gy gamma ray / week for the last 8 weeks) for 15 weeks. The results showed that DMH injection induced colon polyps, biochemical changes such as increased CEA, COX-2, oxidative stress, kidney function tests with the reduction of antioxidants markers. Also histopathological changes in both kidney and colon tissues. While exposure to gamma rays or oral administration of aspirin and LUT either alone or combined showed a significant decrease in CEA, COX-2 , oxidative stress and increase the antioxidant enzymes activities. Also they diminished most of the histopathological alterations. In conclusion the combination of aspirin and LUT with low dose gamma irradiation has a synergistic effect and a chemoprevention effect against colon cancer. |
