Abstract Aim of the work:- The aim of this study is to demonstrate the comparative efficacy and tolerability of 3 different progesterone formulations used for Luteal phase and early pregnancy support during assisted reproduction :- 1- Progesterone vaginal gel (Crinone 8%). 2- Progesterone vaginal suppositories. 3- Progesterone intramuscular injection. In order to establish the optimal formula and its route of administration for this purpose. a) To foretell the future management of patients with advanced east cancer depending on sound scientific oncological ormations rather than on emperical clinical trials ,a prospective in forty patients with advanced breast cancer. s study comprises clinical study including historY,examination and breasts,staging,investigations,histopathological biochemical assays for determination of estrogen receptors and protein estimation of biopsied Detenn1nation of estrogen and progesterone receptors in the specimens incl udes the following st eps : ) Tissue storage in minus 700 Cf Freezer after trimming of fat and debris,transport on icetto laboratory and freezing in liquid nitrogen. Preparation of cytosol by pulverization of frozen tissue, addition of suitable buffer(phosphate for progesterone receptors and TED buffer for estrogen receptors),homogenization and centrifugation. samples are prepared as follows: (JH )-17 B¬estradiol 104 Ci/riM for estrogen receptors or (JH )R5020 55.4 Ci/clu is added at 2 X 10-8M(final concentration in 2 microliter ethanol to 250 microliters diluted cytosol ,incubated’ for overnight at 4°C. Parallel samples are preincubated for 10 minutes with a 100 fold excess of unlabeled 17 B-estradiol. |
