Abstract

Instrumental analysis of certain gastro intestinal tract drugs”
The thesis is concerned with the analysis of some drugs acting on gastrointestinal tract through development of sensitive, precise and accurate methods for the determination of the investigated drugs either in the pure form, in their pharmaceutical formulations or in the presence of some drug degradation products with low cost depending on the availability of chemicals and equipments. The investigation includes usefulness of spectrophotometer and HPLC.
These drugs include Albendazole, Secnidazole, Pipoxolan, Bisacodyl, Amoxicillin and Omeprazole.
The thesis consists of three main parts.
Part I
This part is concerned with the demonstration of chemical structure, physical and pharmacological properties of the studied drugs and also a theoretical basic background review on the previous methods of the assay of the cited drugs.
Part II
This part describes spectrophotometric methods and it is subdivided into following parts:
Part II.A:
This part enumerates the reagents and dyes used throughout the spectrophotometric methods, their molecular structures and their utilization as chromogens and also shows the concentration of their working solutions.
Part II.B:
This part describes spectrophotometric methods for the determination of albendazole and secnidazole by vanillin method. The reaction products were measured at λmax 562 nm and 557 nm . The proposed method was applied for determination of the cited drugs in some pharmaceutical preparations. The obtained results were compared with those of the reference methods.
Part II.C:
This part is concerned with determination of albendazole and secnidazole by NQS method. The method describes the nucleophilic substitution of the cited drugs with 1,2-naphthoquinone sulphonate sodium salt. The reaction product was measured at λmax 482 nm and 480. The method was satisfactory applied for the determination of the drugs in tablets and the results were compared statistically with reference methods.
Part II.D:
This part demonstrates determination of albendazole, secnidazole, pipoxolan and bisacodyl by TCNE methods. The method is based on the charge transfer (CT) complexes reaction of the drug with tetracyanoethylene. The reaction products were measured at λmax 395 nm,397 nm,398 nm and 398 nm respectively. The method was satisfactory applied for the determination of the drugs in tablets. Results obtained showed good agreement with those obtained by the reference methods.
Part II.E:
This part describes determination of albendazole, secnidazole, pipoxolan and bisacodyl with bromatometric method using thymol blue, methyl orange and rhodamine B as dyes. The absorbances were measured at λmax 545 nm, 510 nm and 555 nm, respectively. The proposed methods were applied for determination of the cited drug in some pharmaceutical preparations. The obtained results were compared with those of the reference methods.
Part III:
In this part, an isocratic RP-HPLC method has been developed for simultaneous separation and determination of mixture of secnidazole, omeprazole and amoxicillin in their pure forms and pharmaceutical preparations. chromatographic conditions indicate the suitability of the system for analysis of this mixture. The separation was carried out on ACE 5 C18 (25 cm x 4.6 mm ) column using a mobile phase of acetonitrile:potassium dihydrogen phosphate buffer (pH 7.5) (60: 40, v/v) at ambient temperature. The flow rate was 1.5 ml/min and the detection was set at 230 nm. The method was satisfactory applied for the determination of drugs in tablet and capsule the, results were compared statistically with reference methods.
