Abstract

The aim of this study was to evaluate the effect of the finasteride (a potent specific inhibitor of 5α-reductase) on some immunological(IL-6 and TNF-alpha), physiological (Testosterone, glucose, liver function and lipid profile ) , reproductive (sperm count, testes weight and epididymis weight ) and histopathological (testes and liver) parameters. Allium sativum (AS) and Nigella sativa (NS) are used as a treatment to the adverse effects of finasteride . Oral administration of finasteride (a dose of 200 mg/kg body weight/day for 3 months) to male albino rats was followed by oral administration of AS and NS (a dose of 200 mg/kg body weight/day for 45 days). Fourty eight adult male albino rats Rattus rattus weighting 150-200 gm, were used in the present study. Animals were classified into 6 equal groups each comprising eight ones as follows: I- Normal group, II- Control group (AS+NS), III- Finasteride group, IV- Finasteride, AS group, V- Finasteride NS group, VI- Finasteride, AS and NS group. The finasteride drug has a negative effects on the immunological parameters and some hepatic functions and leads to increasing in the glucose and bad lipids while reduce the fertility parameters and the histopathological effects the testis and liver tissues are highly destructed by finasteride administration. In contrast NS and AS corrected some of these negative effects by using each of them separately or in combination together. where there was a great reduction in IL-6 and TNF-alpha levels in the positive control group and the finasteride group also a reduction in the levels of the finasteride treated group with NS and finasteride treated group with As and finasteride treated group with both of NS & AS together in comparison to the control group. Hormonal assay showed a reduction in testosterone levels in the control group and in the finasteride treated group with AS while the levels increased in finasteride group, finasteride treated group with NS and finasteride treated group with both of NS & AS together. Also, liver functions shows, A great reduction in protein levels in the control group and the finasteride group also a reduction in the levels of the finasteride treated group with NS and finasteride treated group with As and finasteride treated group with both of NS & AS together. A significant reduction in ALT levels in the control group, finasteride group and finasteride treated group with As and finasteride treated group with both of NS & AS together while the levels were increased in the finasteride treated group with NS. A significant decreasing in AST levels in the control group, the finasteride treated group with NS and finasteride treated group with As and finasteride treated group with both of NS & AS together while the levels where increased in the finasteride group. Also, a decreasing in ALP levels in the control group and finasteride treated group with both of NS & AS together while the levels increased in the finasteride treated group with NS and finasteride treated group with As and finasteride treated group. A significant decreasing in Glucose levels in the control group and in the finasteride treated group with NS and finasteride treated group with As and finasteride treated group while the levels increased in finasteride group and finasteride treated group with both of NS & AS together. The lipid profile results demonstrated a reduction in cholesterol levels in the positive control group and in the finasteride treated group with AS while the levels increased in finasteride group, finasteride treated group with NS and finasteride treated group with both of NS & AS together. A highly decreasing in the levels of triglycerides in the positive control group, the finasteride treated group with AS and finasteride treated group with both of NS & AS while the levels increased in finasteride group, finasteride treated group. A highly increasing in the levels of HDL in the positive control group, finasteride treated group with NS, the finasteride treated group with AS and finasteride treated group with both of NS & AS while the levels decreased in finasteride. A highly decreasing in the levels of LDL in the positive control group, finasteride treated group with NS, the finasteride treated group with AS and finasteride treated group with both of NS & AS while the levels increased in finasteride group. The fertility results showed a highly increasing in the count of sperms in the positive control group, finasteride treated group with NS and finasteride treated group with both of NS & AS while the count decreased in finasteride group and the finasteride treated group with AS . An increasing in the weights of testes in the positive control group while the weights were decreased in finasteride group, the finasteride treated group with AS, finasteride treated group with NS and finasteride treated group with both of NS & AS. An increasing in the weights of epididymis in the positive control group while the weights were decreased in finasteride group, the finasteride treated group with AS, finasteride treated group with NS and finasteride treated group with both of NS & AS in comparison to the control group. The histopathological results appeared Finasteride (FIN) administration induced several histopathological alteration in the testis where FIN degenerated and reduced the cellular layers inside the seminifrous tubules but increased the number of leydig cells. The use of NS and AS after finasteride administration helped the seminiferous tubules of testis to restore their normal histological structure. Also, Finasteride (FIN) administration and AS treatment in high dose induced several histopathological alteration in the liver where FIN degenerated and caused necrosis to the hepatocytes and other hepatic components While NS treatment restore the normal histological structure of the liver..So, it is preferred to use these two herbs with the drug administration to avoid its side effects.
