Abstract

Nicotine is a combination of pyridine and methyl
pyrolidine, C10H14N2. This study was aimed to investigate
the biochemical and histological effects of nicotine on testis
of adult rats.
Nicotine caused a significant malfunction (P < 0.05)
in the mean values of serum sexual hormones, testosterone,
ABP, FSH and LH concentration in the testis when
compared with the control. It is concluded that nicotine
caused decrease of testosterone and increase of ABP, FSH
and LH concentrations in the serum. Also it causes
histological changes in the testis and theses changes were dose dependndant.
A total of adult 30 male rats weighing from (250-300)
gm were divided into three groups: Group 1 (n=10), the
control group was given normal saline1m subcutaneous pear
day for 8 weak ; group 2 (n=10), was given nicotine at low
dose ”0.25mg/kg” body weight;/ day/8weak, group 3
(n=10), was given nicotine at high dose 0.5mg/kg” body
weight.
After sacrifice, blood samples were collected in tubes,
separated the serum for determination of testosterone,
androgen binding protein (ABP), FSH and L.H.
The results showed that serum testosterone levels
were significantly decreased in both treated rats groups, low
and high dose, compared with control animals (P < 0.05). on
the other hand, serum androgen binding protein (ABP),
follicle stimulating hormone (FSH) and luteinizing hormone
(LH) levels were significantly increased in both treated rats
groups, low and high dose, compared with control animals
(P < 0.05).
Histological results revealed destruction changes in
rates testis of low dose nicotine-treated group and this
changes were increased with high dose –treated group.
this study aimed to determining the biochemical effect
of nicotine on male testis by evaluating, serum
concentration of testosterone, androgen binding protein
(ABP), FSH and LH. So . Nicotine should therefore be
taken with caution to avoid its harmful effects on testis.
