Abstract

Salmonellosis is a zoonotic disease. More than 2500 serotypes, mostly belonging to Salmonella enterica. This study sought to determine wither Salmonella Enteritidis isolates obtained from forming egg associated with colonize reproductive tissue and deposited inside eggs laid by naturally infected hens had unique DNA alles or notA total number of 90 layer hens from different farms (30 from farm No. one, 25 from farm No. two and 35 from farm No. three) were collected for bacteriological examination (Delayed secondary enrichment) and PCR. It showed that 8 layer hens out of 90 with percentage (8.88%) were positive for isolation of Salmonella Enteritidis and atotal number of 145 of its eggs were colected for bacteriological examination These isolates were subjected to serological identification which carried out at Animal Health Research Institution (AHRI) by using of polyvalent and monovelant O and H Salmonella antisera and confirmed by Multiplex PCR technique which carried out at faculty of veterinary medicine, kafer EL- Shekh University as Multiplex PCR is a technique for in vitro amplification of specific DNA segments. It is extremely sensitive allowing the detection of minimal quantities of target DNA, so considered a good tool for diagnosis. Lastly we confirmed that the hot strain infected ovary, intestine and forming egg of naturally infected one layer hen had a unique DNA alles by ERIC- PCR which carried out at faculty of veterinary medicine, kafer EL- Shekh University The incidence of Salmonella Enteritidis isolated from laying hens reproductive tissue were 5, 2 and 1 from ovary, oviduct and forming eggs respectively with percentage 5.55%. 2.22% and 1.11% respectively While the incidence of Salmonella Enteritidis isolated from intestine of laying hens were 7 with percentage of (77.7%). More over the incidence of Salmonella Enteritidis isolated from outer shell of egg laid were 2 with percentage of 1.37% and from internal egg laid contents were 1 with a percentage of 0.68%. There is one case Salmonella Enteritidis isolated from ovary and not detected in intestinal tissue which may be due to ascending infection from cloacae and/or recovery of the hen due to treatment There is also one case of Salmonella Enteritidis isolated from ovary, intestine and forming egg and this case examined to detect the genome pattern of the micro-organism that confirmed by ERIC- PCR that Salmonella Enteritidis isolates of this case had a unique genome pattern This study showed that the deposition of Salmonella Enteritidis inside developing eggs is a direct consequence of reproductive tissue colonization in systems of naturally infected layer hens and the isolated Salmonella Enteritidis from formed eggs, ovaries and intestine had unique genome pattern which indicated that this serotype of Salmonella Enteritidis has intrinsic characteristics that allow it to survive the attacks with the help of antimicrobial molecules during formation of the egg in the hens oviduct and inside egg. This needs more investigation.
