Abstract

The establishment stage with nodal explants of 1cm length and 2-3 mm. thickness obtained from growing Nemaguard peach rootstock seedlings of 2 years old proved the efficiency of 2ppm BA in MS medium for shoots proliferation.During multiplication stage for the shoots of 1cm length obtained from establishment stage , different concentrations as 1,2,4 or 6ppm of BA or Kin. in MS medium indicated the efficiency of 4ppm BA in producing high number of shoots (5- 8.5 shoots/ explant) and number of leaves / shoot. NAA and BA at 0,1,2 and 4ppm each alone or in combinations in MS medium declared that NAA supressed multiplication and produced roots either NAA alone or in combination with BA and 4ppm BA alone still the best for multiplication. Adenine sulphate at 10, 20 or 40 ppm alone or combined with 1, 2 or 4ppm BA did not show benefical effect of adenine sulphate in enhancing more multiplication. BA at 1,2 or 4ppm and diphenyleurea at 0.25, 0.50 or 1ppm each alone or in combinations did not prove efficiency in multiplication compared with 4 ppm BA alone. Also BA at 4ppm alone or combined with 2,3,5- triiodobenzoic acid (TIBA) at 0.50, 1, 2 or 4 ppm did not prove efficiency in multiplication as using 4 ppm BA alone. MS medium the efficious medium comparing to McCown woodyplant medium or B5 medium when amended with 4ppm BA in multiplication. Also adjusting pH to 4 or 5.8 in MS medium was the effective than pH 6.5 concerning multiplication and solidification . MS medium with agar at 6, 8 or 10g/l did not show differences in multiplication . Addition of peptone at 1, 2 or 4 g/l to MS medium supplemented with 4ppm BA did not show efficiency in multiplication rate comparing to MS medium amended with 4ppm BA alone.During rooting stage, NAA, IBA or IAA at 0, 1, 2 or 4ppm in MS medium ,all those auxins resulted in shoot rooting, but the best rooting percentage and number of roots or root length belonged to 2ppm IBA in full MS medium comparing to 1/2 or 1/4 MS strength. Adding activated charcoal to full or 1/2 MS medium and 2ppm IBA decreased rooting process. Using Mccown woody plant medium as full or 1/2 strength either alone or amended with IBA at 2ppm and 2,3,5- triiodobenzoic acid (TIBA) at 0.5 ppm, declared that TIBA supressed rooting process.The obtained rooted plantlets could be acclimatized in peat moss alone, 1sand + 1peat moss, 2 sand + 1 peat moss or 3 sand + 1 peat moss (v/v) successfully and transplanted to soil by 100% survival percentage. Peat moss alone was the best medium for this stage.
