Abstract In the present study, male albino rats were randomly divided into 6 groups; control (untreated), animals treated with diacerein(50 mg/kg), diacerein(100mg/kg),cisplatin (5mg/kg), cisplatin+diacerein(50 mg/kg),and cisplatin+diacerein(100mg/kg). Administration of diacerein was started 11 days before cisplatin administration and continued for further 3days after cisplatin administration. br Results showed that kidney/body weight ratios in either diacerein (50 mg/kg) or diacerein (100mg/kg) groups did not significantly differ -#102;-#114;-#111;-#109; that of control. On the other hand, kidney/body weight ratios in cisplatin group significantly increased compared to control group. Pre-treatment with either diacerein (50 mg/kg) or diacerein (100mg/kg) before cisplatin did not significantly decrease the kidney/body weight ratio. br Administration of a single dose of cisplatin (5mg/kg, i.p) caused significant deterioration in renal function, designated by the increase in serum levels of both urea and creatinine as compared with control group. Pretreatment with either diacerein (50 mg/kg) or diacerein (100mg/kg) before cisplatin caused a significant decrease in serum levels of both urea and creatinine as compared with their respective cisplatin treated group. br In urine samples -#102;-#114;-#111;-#109; cisplatin treated groups, there is significant increase in microalbumin level and sodium level and decrease in 24h urine volume and its creatinine level. Pre-treatment with either diacerein (50 mg/kg) or diacerein (100mg/kg) before cisplatin caused a significant decrease in microalbumin level and sodium level and increase in 24h urine volume and its creatinine level. br In renal samples -#102;-#114;-#111;-#109; cisplatin treated group, cisplatin caused oxidative stress, as evident by the significant decrease in renal GSH and catalase. Pre-treatment with either diacerein (50 mg/kg) or diacerein (100mg/kg) before cisplatin significantly reversed the oxidative effect of cisplatin on renal GSH and catalase. br Diacerein also significantly decreased renal MDA compared to control group, which was reversed by pre-treatment with either diacerein (50 mg/kg) or diacerein (100mg/kg) before cisplatin administration. |
