Abstract The present work aimed to study the developmental changes in the histological and histochemical structure of the liver in albino rats under the effect of methotrexate and methotrexate with hydrocortisone by both light and scanning electron microscope. Fifty albino rat embryos and 105 of both sex rats at different ages and weights were used in this study and classified as follows: 1- A control group included ten embryos at the 14th and 20th day and fifteen rats of both sex at two weeks, one month and adult for investigation of their livers. 2- An experimental group included forty embryos at the 14th and 20th days and 90 rats of both sex at two weeks, one month and adult for studying the effect of methotrexate and methotrexate with hydrocortisone on the liver of these animals. Every prenatal and postnatal liver was divided into two parts. One part was fixed in Karnovsky’s fixative and cut into slices to study the morphology of the structures seen in the cut surface by using the scanning electron microscope. The other part was divided into many parts and put in formal saline, formal calcium and Carnoy’s fixative and stained by haematoxylin and eosin, Mallory’s trichrome, PAS, Gomori’s method for alkaline phosphatase and methyl green pyronin stains to demonstrate the general structure, connective tissue, PAS-positive material, alkaline phosphatase enzyme and nucleic acids respectively. |
