Abstract

The aim of the present study to investigate the hypoglycemic, hypolipidemic and antioxidant activities of ethanolic extract of Purslane and Sonchus Asper in streptozotocin (STZ)-induced diabetic rat. Acute toxicity was studied in rats after the oral administration of Purslane and Sonchus Asper to determine the dose to assess hypoglycemic activity. The median lethal doses (LD50s) of Purslane and Sonchus Asper in rats were 2880 and 4420 mg/Kg b.w., respectively. In rats, Purslane and Sonchus Asper extracts were orally given for 56 days and its effect on body weight, plasma glucose, serum insulin, total cholesterol, HDL - cholesterol, LDL- cholesterol, triacylglycerol , Alanine amino transferase (ALT), Aspartate amino transferase (AST) activities, total protein, albumin and globulin, albumin/globulin ratio, creatinine, were determined every 2 weeks (14, 28, 42 and 56 day). Also liver, kidney and heart malonodialdehyde (MDA), reduced glutathione (GSH) and super oxide dismutase (SOD) activity were determined at the end of the experiment. Histopathological examination of the liver, kidney and heart was determined at the end of the experiment. Oral daily administration of both the whole parts of Purslane and Sonchus Asper extracts at a concentration of 288 and 442 mg/kg b.w, respectively, showed a significant decrease in plasma blood glucose, total cholesterol, LDL- cholesterol, triacylglycerol, ALT, AST activities, creatinine and liver, kidney and heart MDA. The treatment also resulted in a significant increase in serum insulin, HDL-cholesterol, total protein, albumin, globulin and albumin/globulin ratio and level of reduced glutathione and superoxide dismutase activity in the liver, kidney and heart of diabetic rats and also showed a protective effect in the structure of the liver, kidney and heart. The results clearly suggested that, the whole parts of Purslane and Sonchus Asper treated groups may effectively normalize the impaired antioxidant status in Streptozotocin - induced diabetes. The extracts exerted rapid protective effects against hyperglycemic, hyperlipidmic effect and lipid peroxidation by scavenging of free radicals by reducing the risk of diabetic complications.
