Abstract

The spread of viral diseases in Egypt during the last decade had enormously impacted on the animal productivity, milk products and human public health. Therefore, the present study aimed to the detection of Rota, Corona and foot and mouth disease viruses in raw milk and milk products (cheese and yoghurt). This study included of 100 raw milk samples collected randomly at El-Gharbia Governorate markets from October 2011 till June 2012 for detection of rota and corona virus antigens by ELISA. The antigen of rota virus could be detected in milk samples by the percentage of 15% and corona virus by 5%. The highest percentage of rota in El-Mehalla El-Kobra district and the corona virus in couttor district. The study included 60 samples of different types of cheese from the same examined area for detection of rota and corona viruses antigen by ELISA. The antigen of rota and corona viruses in kareish cheese could be detected in (5) and (2) respectively out of (30) samples. The antigen of rota and corona viruses in unpackaged soft cheese could be detected in (2) and (1) respectively out of (20) samples . The antigen of rota and corona viruses could not be detected in packaged soft cheese. The study included 50 samples of different types of yoghurt collected randomly, from the same examined area for detection of rota and corona viruses antigen by ELISA. The rota virus antigen in zabadi could be detected in (3) samples out of (40) and the corona virus antigen could not be detected in zabadi, and the results indicate that the plain yoghurt were free from rota and corona virus antigens. This study also included 100 serum samples and 100 whey samples and 30 tongue epithelial tissues and ruptured vesicles samples from suspected dairy animals with FMD from February till June 2012 for detection of FMD virus antigen and antibodies. Nonstructural protein (NSP) against FMD virus in serum samples could be detected in (87) out of (100) samples by prioCHECK FMD virus (NS), and the highest percentage in Gouttor district. The antibodies of FMD virus in whey samples could be detected in 50% type A, 63% type O and 75% type SAT2 by liquid phase blocking ELISA . FMD virus could be isolated from epithelial tissue and ruptured vesicles by inoculation on cell culture Baby Hamester kidney cell 21 (BHK21) and the cytopathic effects (CPE) could be induced in (5) samples out of (30) samples after 72 hours. The antigen of FMD virus in the same examined (30) epithelial tissue samples could be detected in (4) type A, (2) type O and (10) type SAT2 by indirect sandwich ELISA. The public health hazard of rota, corona and foot and mouth disease viruses and methods used to control spreading of disease among animals and human were discussed.
