Abstract

A light microscopic study was done on the postnatal development
of the white pulp of the spleen and the cortex of mesenteric lymph
nodes in the albino rat, using H & E and immunoperoxidase staining
techniques. In the latter technique, monoclonal antibodies were used
for identification of T and B lymphocytes. The white pulp in most of
the spleens of seven-day old rats consisted of a mixture of mainly small
lymphocytes with some medium-sized lymphocytes surrounding the
central arterioles and forming a primitive PALS. No definite marginal
zone could be detected. The majority of T cells were small lymphocytes
and were concentrated around the central arterioles. B cells,
most!)( in the form of medium-sized lymphocytes. were located
peripherally-fin twenty-one-day old rats, the white pulp consisted of 2
zones, the PALS which is divided into a larger central area densely
populated with small T lymphocytes and a smaller peripheral area
containing mainly small lymphocytes and occupied by both lymphocyte
populations, and the marginal zone, separating the PALS from the red
pulp, which contained a large population of medium-sized B lymphocytes.
Primary follicles were infrequently seen.; However,
follicles formed of aggregated small B lymphocytes, as
primary
well as
secondary follicles with pale germinal centers were observed in all
