Abstract

Analytical study of some drugs used in psychiatric disorders
with antidepressant effect of forensic medicine interest
Lately, psychiatric drugs have found wide use among large sectors of young as well as
old patients. Accordingly, it is deemed necessary to propose suitable methods to assay
these drugs in their pharmaceutical preparations and in biological fluids.
The thesis compromises three main parts:
Part I
This part contains a short account about psychiatric disorders and different categories of
anti-psychotics. Also the pharmacological actions and side effects of each category are
discussed. A representative example of two drugs (Aripiprazole (ARP) and Quetiapine
(QTI)) under study is given with a highlight on their chemical structures, formulae,
chemical names, physical properties and pharmacological effects.
Furthermore, it includes literature reviews for the reported methods for the selected
drugs in pharmaceutical dosage forms as well as biological fluids.
Part II
This part deals with different analytical techniques for the determination of ARP in
tablet dosage forms and human plasma. It compromises two chapters:
Chapter 1:
In this chapter, two spectrophotometric methods have been described for the
determination of ARP in tablets. The first method was based on measurement of the
absolute values of 1D amplitudes at 225nm and of peak to peak at 245 and 268nm. 1D225
and1D245-268 in methanol gave linear calibration curves at concentration ranges of
0.3-1 µg/ml for 1D225 and 2-6µg/ml for 1D245-268.
The second method was based on the formation of an ion pair complex between the
drug and Alizarin Yellow G and the absorbance of the highly yellow colored product was
measured and quantified spectrophotometricaly at 490nm without any extraction step. This
complex is formed through the interaction of the protonated form of the basic tertiary
amino group of aripiprazole and the carboxylate anion of AYG in an acidic medium of
phosphate buffer pH 4.2. Factors affecting color development were studied and optimized
for the reaction.
