Abstract In the in vitro studies, it was found that both UCBSCs and BMSCs with their two types MSCs and HSCs could be proliferated and differentiated into hepatocytes in vitro under stress of certain conditions. MSCs and HSCs separated from human umbilical cord blood samples and from male mice bone marrow samples yielded 2 x 105 cells/ml blood sample. MSCs and HSCs of both UCBSCs and BMSCs were proliferated in primary culture. After addition of mouse hepatocyte extract, they significantly underwent a differentiative stage appearing as rounded hepatocyte-like cells. In the in vivo studies, the effect of UCBSCs and BMSCs- with their two types MSCs and HSCs- transplantation on treating CCl4- induced liver fibrosis of their mice groups was investigated in order to study stem cells differentiation into hepatocytes. It was found that both UCBSCs and BMSCs with their two types showed a hepatoprotective and therapeutic effect against the CCl4-mediated liver injury indicating the differentiation into hepatocytes. The biochemical, histological and molecular changes induced by CCl4 were improved. |
