Abstract

This research was carried out to evaluate the hypolipidemic effect and to investigate the essential changes produced by three lipid-lowering drugs (Gemfibrozil , Fluvastatin and Sirtvastutin) in addition to kidney functions and transaminase activities.
One hundred and eight male albino rats were divided into two main groups according to the type of diet served: FIRST GROUP :
Consists of 12 rats fed on a normal diet . SECOND GROUP:
Consists of 96 rats fed on hyperlipidemic diet which contains cholic acid, cholesterol and thiouracil for 45 days and continued throughout the drug administration and divided intc two groups. Group (A) :
This group consists of 24 rats and received normal saline and used as control which divided into two subgroups: Subgroup (Al) :
Consists of 12 rats and administrated saline for 30 days. Subgroup(A2):
Consists of 12 rats and administrated saline for 45 days. Group (B) :
This group divided. into three subgroups according to the type of drug . Subgroup (1):
This subgroup consists of 24 rats and administrated one dose daily from Gemfibrozil by stomach tube according to the therapeutic dose and divided into :
Subgroup (la):
Consists of 12 rats and administrated the drug for 30
days
Subgroup of 12 rats and administrated the drug for 45
days. Subgroup (2):
This subgroup consists of 24 rats and administrated one
dose daily from Fluvastatin by stomach A.ube according to the therapeutic dose and divided into :
Subgroup (2a):
Consists of 12 rats and administrated the drug for 30
days
Subgroup (2b):
Consists of 12 rats and administrated the drug for 45
days. Subgroup (3):
This subgroup consists of 24 rats and administrated one
dose daily from Simvastatin by stomach tube according to the therapeutic dose and divided into :
Subgroup (3a):
Consists of 12 rats and administrated the drug for 30
days.
Subgroup (3b):
Consists of 12 rats and administrated the drug for 45
days.
Blood samples were collected in the morning at the end
of each period where the rats are in the fasting state and the serum was separated to estimate the following parameters :
Total lipids, Cholesterol , Triacylglycerol , LDL Cholesterol , HDL Cholesterol , Phospholipids , NEFA concentrations , Lipase and LCAT activities in addition to kidney functions ( urea and creatinine) and transaminase activities (AST & ALT ).
The results showed the effect of hyperlipidemic diet on normal rats include a highly significant increase in the levels of serum Total lipids , cholesterol , triacylglycerol , LDL-C , HDL-C , phospholipids concentrations and LCAT activity but there were a highly significant decrease in the levels of NEFA concentration and Lipase activity . The side effect of hyperlipidemic diet showed a highly significant increase of serum ALT activity and urea ,while there was a highly significant decrease in serum creatinine but no
change in case of AST activity..
Mier administrating the drugs ,the results showed a
highly significant decrease in the levels of serum total lipids , cholesterol , triacylglycerol , LDL-C , HDL-C , phospholipids and LCAT activity but there was a highly significant increase in the levels of NEFA and Lipase activity in the 30 days treatment period .The more effective hypolipidemic drug in this period
was Gemfibrozil.
The side effect of the drugs administrated showed a
highly significant increase of serum AST , ALT activities and urea but there was a highly significant decrease of serum creatinine .The less effective drug was Fluvastatin through 30
days treatment period.
In the 45 days treatment period , the results showed a
highly significant decrease in the levels of serum Total lipids , cholesterol , triacylglycerol , LDL-C , phospholipids and LCAT activity but there were a highly significant increase in the levels of NEFA and Lipase activity , there was a highly significant decrease of serum HDL-C in Gemfibrozil but increase in both of Simvastatin and Fluvastatin in the 45 days treatment period . The more effective hypolipidemic drug in this period was
Simvastatin.
The side effect of the
highly significant increase of concentration but there was a serum creatinine which tetumcd The less effective drug was treatment period.
