Abstract The oral LD50 of Basagran in Agoza strain tQmale rats was found to be (380 mg/kg B,W~)0 The clinical symptoms which appears on animals during determination of T~D50 and these administered 350 mg/K.g B.W. at 18 days of gesta— tion were skin rash on nose, mouth, eyes, swelling at the maxilla of fore limb on the right side and tremors all over the body. Organs from animals died during the determination were preserved in 10% formaline and histopatholc’gical examination were carried out. Examination revealed presence of telangectasis in liver with coagulative necrosis of the hepatic cell. Lung showed nedema in the perivascular areas and air alveoli lymphoid cell hyperplasia were noticed. cloudy swelling and coagulative necrosis in kidney. Isohemic neural damage of purkinge cells in brain, diffuse gliosis in the white matter and perivascular gliosis were noticed. Distribution of Basagran in different organs was determined (in a group received 380 mg.Kg B.W by combined thin layer chrnniatrgraphic technique and ultra violet spectrnphotnmeteric analysis. The effect of Basagran on pregnant mothers, foeti placenta and embryones were investigated through intragastric intubaction with 350 mg,ICg.B.W.dose of Basagran at 18 days of gestation period to 18 animals, Cesarian sections were carried out en 18 animals after 24 hours pest administration foeti, umbilical cord, placenta and mother organs were subjected to histo— pathological examination results showed the same pre vious his tcpatholc’gical lesion, The most important colour tests which gave posi tive results with Basagran were vitalis test (bloody red colour) Isieberman’s test (yellow lemon colour), Frohad’s test (violet colour) and mandelin’s (red colour) The RF values of Basagran on silica gel using mixture of some aromatic hydrocarbones were also determined. The spectrophotometeric analysis showed that Basagran has an absorption maxima at 270 nm wavelength. Thin layer chro’matographic analysis provide the presence of Basagran in embryonic tissues—extract. Teratological effect of Basagran were investi— gajed on sixty pregnant Agoza strain female rats, classified into 5 groups (each 12 animals) A, B, C, D, E and each group was classified into four sub groups each contains 3 animals (A1, A2, A3, A4, B1, B2, B3 etc). A single oral administ ration of herbicide was carried out at different period of gestation (6, 8, 11, 14, 16). Three doses (25, 90, 200 -mg/Kg B0W.) were used at each gestation interval. The teratolr’gical studies showed that: 1. The resorption of freti were high in groups administered the herbicide at 6, 8 days of gestation (75%-60%). Then gradually lowered in groups dosed at 11, 14, 16 days of gestation. 2. The size of the foeti were smaller than centre]., Absence of coccygeal vertebra.e was recorded in all groups, Metacarpal, metatarsal phalanges were absent in all groups except in group dosed at 16th days of gestation. |
