Abstract The results of the present study show that acute endotoxemia in rats is associated with cardiac, hepatic and renal dysfunction. Elevations of serum markers of cardiac (CK-MB and LDH), hepatic (ALT and AST) and renal (creatinine and proteinuria) dysfunction and inflammation (NO and CRP) were demonstrated after LPS injection in rats. Inhibition of iNOS by pretreatment with the highly specific and potent iNOS inhibitor 1400W prevented these LPS-induced elevations in serum markers of multiple organ dysfunction. Similarly, inhibition of NO signaling through pretreatment with the sGC inhibitor ODQ prevented this LPS-induced organ dysfunction. Conversely, pretreatment with the powerful anti-inflammatory and immunosuppressant steroid dexamethasone was unable to prevent LPS-induced organ dysfunction. These results confirm that multiple organ dysfunction associating acute endotoxemia in rats can be prevented by blocking excessive NO production through iNOS inhibition or by blocking NO signaling through sGC inhibition, but not by using corticosteroids in large dose. |
