Abstract

(I) This work was planed to study the pattern of toxoplasmosis in cats and mice and its seroprevalence in sheep and goats in Sharkia province.
(2) Fifty seven apparent normal cats 2—3 months old and 171 mice were selected for this study.
(3) Toxoplasma gondii RH strain (tissue cysts) was kindly obtained as brain emulsion for experimental infection.
(4) Three cats were infected orally with the obtained tissue cysts for obtaining sporulated oocysts.
(5) Three mice were inoculated I/P with the obtained tissue cysts for obtaining tachyzoites.
(6) Twenty four cats were divided into 4 groups to study the disease pattern:
a) 1St group 10 cats fed V2, 1, 2, 3 and 5 ml of the brain
emulsion containing tissue cysts, 2 cats for each dose.
b) 2’~” group 6 cats infected orally with 60.000 and 120.000
sporulated oocysts, 3 cats for each dose.
c) 3”~ group 5 cats were fed 5 mice containing tachyzoites
(infected three days ago).
d) 4th group 3 cats were kept as control.
e) Observation of cats and examination of their feces were
conducted daily up-to 6 weeks.
f) Blood samples collected weekly from experimentally
infected and control cats up-to 6 weeks.
g) Histopathological examination and bioassay were applied
from tissues of killed cats.
(7) The results revealed that:
a) Clinical txoplasmosis not observed in the 2’~’ and 31~1 groups while the group of cats infected orally with tissue cysts had different symptoms as diarrhea, vomiting and nervous signs and 4 of them were reached the state of acute
toxoplasmosis.
b) All cats in the l~ group were shed T.gondii oocysts, while
only 4/6 and 4/5 cats in the 2’~ and 3”’ groups were shed
T.gondii oocysts.
c) Bioassay procedure is more sensitive in detection of
T.gondii infection in tissues than histopathological examination.
d) LAT was neither sensitive nor specific, while ELISA was
very sensitive and specific in detection of antitoxoplasma
antibodies in infected cats.
(8) 24 mice were divided inti 4 equal groups:
a) 1st group inculated I/P with tissue cysts of toxoplasma RH
strain (1/2 ml for each mouse).
b) 2nd group inoculated I/P with 60.000 sporulated oocysts.
c) 3”’ group inoculated I/P with 4 x 106 tachyzoites.
d) One mouse from each group was killed after 3 days and
examined for presence of T.gondii tachyzoites.
e) Another one mouse from each group was killed weekly for
blood and tissue bioassay and histopathological studies.
(9) The result revealed that:
a) All mice infected with sporulated oocysts were died 14
days post-infection.
b) Bioassay procedure in mice was also more sensitive for
detection of T.gondii than histopathological examination.
c) Tissue bioassay in cats was more sensitive than that used in mice, moreover in cats it can differentiate between the
stages of toxoplasma present in tissue.
(10) Serum samples collected from 364 sheep and goats (275 sheep
and 89 goats) in different localities were examined by LAT for
detection of specific antitoxoplasma antibodies.
(11) The result revealed that:
a) 80 (21.98%) [56 (20.36%) in sheep and 24 (26.97%) in
goatsj were positive with titres ranged .
b) The highest rate of infection in sheep and goats was
recorded in Zagazig city (27.4%), while the lowest rate
was recorded in El-Saihia city (12.7%).
