Abstract

Cucumber is an important raw vegetable crop, sensitive to a wide range of contaminants. Hence, the present study was carried out to evaluate physiological, ultrastructural and molecular responses of cucumber plant and its cultured tissues exposed to essential heavy metals such as zinc, nickel and non essential heavy metal such as cadmium. For induction of cucumber cultured tissues; different explants were cultured on Murashige and Skoog (MS) medium supplemented with auxin combined with cytokinin at different concentrations. MS medium supplemented with 0.05 mg/L NAA and 0.5 mg/L BAP proved to be the best medium for initiation and maintenance of callus-derived hypocotyls of cucumber. from physiological view, there were significant variations in the contents of pigment fractions, total soluble protein, carbohydrate, proline, phenolic and antioxidant enzymes (catalase, polyphenol oxidase and peroxidase) in the treated cucumber plant and cultured tissues compared to control
