Abstract

The present study was done to evaluate bacteriological quality of some canned and processed foods through monitoring bacteriological qualityof nine different food products distributed along the markets in Qualubia governorate Egypt .This study was carried out during aperiod of study extended from June 2010 to November 2011.The sampling equipments microbiological and food industry depts. Fac. Agric. Ain shams University. The present study aimed for isolation and identification of foodborne microorganisms and its determination of intrinsic parameters that affected microbial growth in processed foods and comparison between the results obtained and hazard analysis critical control point of food point. Canned and Processed food samples analysed in this study were: Peas, lancheon, tuna, basterma, broad bean, mixed fruit jam, corn, tomatoes, figs jam.The bacterial species which were isolated and identified were: Salmonella , E.coli, Lactobacillus, and Ps.aeruginosa.Identification Process for all species was applied using cultural, physiological and biochemical tests, The highest frequency was found in Ps.aeruginosa isolates, so molecular identification applied on four Ps. aeruginosaisolatesby amplification of 16S rRNA structural gene using PCR,also agarose gel electrophoresis was applied on the isolated 16S rRNA fragment and proved that it was consists of 371bp.By determination of the nucleotide sequences of each isolate, it was found that there were variations in a number of nitrogen bases of each isolates which proved that,the isolated Ps.aeruginosa were strains.Keywords: Identification, Isolation,PCR, Processedfoods,Ps.aeruginosa.
