Abstract Nitroxyl chemistry and biology are studied through donor species and the most well-known donor is Angeli’s salt. Administration of Angeli’s salt to animals results in enhanced myocardial contractility and relaxation. HNO also acts as a potent vasodilator in a mechanism different from NO. These two actions promote the use of HNO in the treatment of congestive heart failure (CHF). Due to the important biological actions of HNO, it is important to develop more donors to aid in the demonstration of the important biological activities of HNO. Based on this goal, this thesis aimed to synthesize new HNO donors that are different from AS. These compounds include acyloxynitroso compounds that release HNO at different rates upon hydrolysis of ester linkages at neutral pH, Diels-Alder adducts that release HNO after oxidation through a cheletropic reaction giving an acyl nitroso intermediate and new phosphinoylhydroxylamines that release HNO under physiological conditions. |
