Abstract This study was designed to Isolate and identify FMD viruses from Suez Canal area governorates. Characterization of VPl gene of FMDV serotypes (0 and A) circulating field strain was carried out and compared with national and international FMD strains. 47 Tongue epithelial and 170 Saliva samples were collected from Suez Canal area. For isolation of FMDV, suspected saliva samples were inoculated in baby mice and BHK 21 cell culture. Out of 167 saliva samples, 48 are positive with 28.74%, while in case of isolation of FMDV suspected T.E samples in baby mice and BHK 21 cell culture 42 samples out of 44 were positive with 95.45%. Using ELISA to detect FMDV, serotype 0 and A were detected in saliva samples by 23.35% and 5.99%; respectively. Whereas, 79.54% and 15.91% of tongue epithelium were positive to serotype 0 and A; respectively. Six PCR product samples were selected as one for FMDV serotype (0) and one for FMDV serotype (A) from each governorate and submitted for performing the sequencing. Results of the sequencing of VPl gene of FMDV serotype (A) revealed that there is no significant divergence between the isolated strains of FMDV type A as the divergence varied from 0.09% to 2.34% among them. Also FMD AlEgypt 2006 isolate is the most nearly isolate to FMD AlIsmailia, FMD AI Suez and FMD AlPort Said. While the results of the sequencing of VPl gene (lD) of FMDV serotype (0) showed high similarity between the isolated strains of FMDV type 0 as the divergence was 0.32% to .97% among them. Also FMD O/Pan Asia isolates is the most nearly isolate to FMD O/Ismailia, FMD 0/ Suez and FMD OlPort Said. |
