Abstract

The study evaluated the immune response elicited by a DNA vaccine encoding mutant phospholipase D protein of Corynebacterium pseudotuberculosis by DNA prime-protein boost protocol. BALB/c mice were respectively vaccinated with plasmid pcDNA-3 encoding mutant phospholipase D protein, with 40-kDa plus recombinant mutant phospholipase D protein, or a combined DNA prime-protein boost regimen. When animals were primed with DNA and boost with 40-kDa plus recombinant mutant phospholipase D protein, both antibodies and cell proliferative response were significantly enhanced. Moreover, this study demonstrated that DNA prime-protein boost inoculation could afforded better protection against an experimental challenge with C. pseudotuberculosis (84%) than those immunized with DNA pime-DNA boost (58%) or those left unimmunized.
