Abstract Several studies have demonstrated that IL-10 have been associated with therapy outcome in haematological malignancies. In addition to expressing full-length IL-10, a new splicing-derived IL-10 variant (termed IL-10δ3) can be detected in ALL samples. It is strongly associated with the good prognosis of acute lymphoblastic leukemia (ALL) in children. Aim of the work: To detect IL-10 and IL-10 isoform, IL-10δ3, expression and its values in childhood BCP- ALL. Patients and methods: samples were collected from 60 pediatric patients with BCP-ALL. The detection of IL-10 and IL-10δ3 were performed by conventional reverse transcriptase polymerase chain reaction (RT-PCR). The results of this study revealed that: • Cases expressing IL-10δ3 showed less incidence of relapse compared to patients expressing only IL-10 and/or IL-10 below detection limit. • cases expressing IL-10δ3 showed more CD34 positivity – with better prognosis- compared to non expressing IL-10δ3. • Immunophenotyping between cases expressing IL-10δ3 compared to non expressing IL-10δ3 was statistically significant. it is rather a predilection to pro B phenotype with IL-10δ3 expression?!. • In the analysis of pEFS, patients with IL-10δ3 expression had a significantly better pEFS at five years (P= 0.047). |
