Abstract

The aim of the present study was to evaluate the effect of administration of selective serotonin re-uptake inhibitor,sertraline ,on reproductive system of rats.
The following items were investigated:-
1-The effect of administration of sertraline on reproductive organs of immature and mature male rats(sex organs weight,histological examination and hormonal assay).

2- The effect of administration of sertraline on placenta and feti of pregnant rats(placental histological examination and fetal skeletal teratology study).
3- The effect of administration of sertraline on suckling male pups(sex organs weight,histological examination and hormonal assay).
Four experiments were carried out:
The first experiment:Effect of sertraline on male rats.

Twenty immature rats (60 days old) were divided into 2 equal groups: Control group (10 rats) were administrated with saline 1ml daily for 115 days.
Treated group(10 rats) were administrated with 10mg/kg sertraline for 115 days.
After that, the body weight and sex organ weights of all groups were recorded and preserved for histological examination .Blood samples were collected,plasma testosterone hormones were estimated.Results showed that administration of sertraline to male rats significantly decreased plasma testosterone, impaired spermatogenic function.

The second experiment:Effect of sertraline on mature male rats.
Twenty mature rats (90 days old) were divided into 2 equal groups:
Control group (10 rats) were administrated with saline 1ml daily for 70 days.
Treated group(10 rats) were administrated with (10mg/kg.b.w.) sertraline for 70 days.
After that, the body weight and sex organ weights of all groups were recorded and preserved for histological examination .Blood samples were collected,plasma testosterone hormones were estimated.Results showed that administration of sertraline in male rats significantly decreased plasma testosterone, impaired spermatogenic function.
The third experiment:Effect of administration of sertraline on pregnancy:
Ten pregnant rats were divided into 2 groups :
1-Control group(5 female rats) administrated with oral saline(1ml) from zero day till day 19 of pregnancy.

2-Treated group(5 female rats) administrated orally with sertraline
(10mg/kg.b.w.) from zero day till day19 of pregnancy.
At day 19 of pergnancy ,the body weight of all rats was recorded.The uterine horns were collected and examined for the presence of black spots which indicates embryonic resorption,the pups and placentas were recorded and placentas were preserved in 10% formalin for the histological study. The morphological changes of feti were noted .Feti were preserved for skeletal teratology study.
Uterine horns taken from sertraline treated dams showed numerous black spots which indicates embryonic resorption ,cause fetal skeletal malformation and placenta of pregnant rats resulted in thickened particularly at labyrinth and basal zones with an increase of mitoses in the including trophoblast cells.
The fourth experiment:Effect of administration of sertraline on suckling male pups:
10 lactating female rats (5pups/dam) divided into 2 groups:
1-Control group; 5 rats were administrated with oral saline (1ml) daily from first day of lactation till weaning day (28 days).

2-Treated group ;5 rats were treated with oral sertraline 10mg/kg.b.w from first day of lactation till weaning day(28 days).
For detection of sertraline effect on pups during lactation , all male individuals were reared until reaching the maturity about 90 days after weaning. After that, the body weight and sex organ weights of all groups were recorded and preserved for histological examination ,blood samples from each rat were collected, plasma testosterone hormones were estimated.Results showed non significant changes in testosterone levels and spermatogenic function.
In conclusion:
1-Selective serotonin reuptake inhibitor(sertraline) in immature and mature male rats may interfere with spermatogenic function with decreased testosterone levels.
2-During pregnancy,sertraline may causes fetal skeletal malformation and induce placental changes as thickening at labyrinth and basal zones with an increase of mitoses in the including trophoblast with hypertrophied nuclei.
3-No effect on reproductive system of daughter male rats of sertraline treated dams during lactation.
