Abstract

Helicobacter pylori is Gram’s negative bacterium that colonizes human gastric mucosa and is one of the most common bacterial pathogens world wide with a prevalence of upto 90 % in developing Countries.This study aimed to is to assess the diagnosis of H.pylori in gastric biopsies taken from patients by using invasive methods, determination of the minimum inhibitory concentrations of some antibiotics by E-test and studying their role in eradication of these isolated H.pylori organisms by culture method after treatment (follow up). Among 50 patients, 44% were positive by culture, 34% were positive by direct Gram’s stain with 77.3% sensitivity, 100% specificity and 90% accuracy.While, 38% were positive by RUT with 63.6% sensitivity, 82.1% specificity and 74% accuracy, and 50% were positive by PCR with 95.5% sensitivity, 89.3% specificity and 92% accuracy with culture as a gold standard. About 90.9% from the 22 isolated H.pylori were sensitive to amoxicillin with MIC value 0.27 µg/ml, 81.8% were sensitive to tetracycline, 54.5% were sensitive to gentamicin, and 90.9% were resistant to metronidazole with MIC value > 256 µg/ml. It was concluded that PCR is the most sensitive method with highly specificity and accuracy for diagnosis of H.pylori in gastric biopsies.While, amoxicillin and tetracycline were suitable antibiotics for eradication of H. pylori infection (P<0.001).
