Abstract

Diabetes mellitus is caused, in most of cases, by diminished rate of secretion of insulin by the beta cells of the langerhan’s for the islets utilization of glucose by the body cells, with a resultant increase in blood glucose, abnormal fat metabolism as well as deposition of lipids in vascular walls to cause atherosclerosis and depletion of protein in body tissues. Other related symptoms are polyphagia and loss of weight. The most important components of diabetic management are diet and insulin The relationship between diet and diabetic is important from the stand point of treatment Aim of study The main objective of present investigation was to study the effect of some commercial herbs and fruit severe hypercholesterolemic diabetic distributed in the local market for treating disorders including serum glucose and cholesterol level so as to show whether they are valid as remedy using experimental animals (rats) experimental design Fourty eight adult male albino rats of spragre Dawley strain 130 -200 were used 44 of them were hypercholesterolemic diabetic, andhypercholesterolemic diabetic rats were the experimental rats remained asnormal healthy ( control group which fed basal diet divided into 12 groups ten of them were previously inflicted with diseases and fed as the followingGroup 1 (4 rats): Basal diet (control negative) • Group 2 (4 rats): Basal diet (control positive) SUMMARY 125 . Group 3 (4 rats): Basal diet + 5% glimepiride>• Group 4 (4 rats): Basal diet + 10% glimepiride.• Group 5 (4 rats): Basal diet + 5% chicory .Group 6 (4 rats): Basal diet + 10% chicory.Group 7 (4 rats): Basal diet + 5% papaya Group 8 (4 rats): Basal diet + 10% papaya. Group 9 (4 rats): Basal diet + 5% Cinnamon.Group 10 (4 rats): Basal diet + 10% cinnamon. Group 11 (4 rats): Basal diet + 5% mixture of herbs.• Group 12 (4 rats): Basal diet + 10% mixture of herbs Blood Samplinge- Blood sampling :from all the previously mentioned groups, blood collected after 12hour fasting at the end of the experiment using the retro-orbital method by means of a microcapillary glass . Blood was collected into a dry clean heparinized centrifuge tubes and a part of blood left to clot in water bath 937c)at room temperature for half an hour, then centrifuged 10 minutesat 3000 r.p.m. to separate the serumSerum was carefully aspirated and transferred into clean quit fitplastic tubes and kept frozen at (-20c) until the time of analysisThe organs (liver, kidney, heart, lungs and spleen)were removedand washed in saline solution, weighted and kept in formalin solution (10,v/v) and sections of fixed liver, kidney, heart prepared according tomethods described by (Drury and Wallington 1980), the relative weighorgans was calculated SUMMARY- 126 Relative weight =weight Animalbodyt Organweigh x 100 The results obtained from this study could be summarized as follow1-Biological results Body weight gain, feed intake and feed efficiency ratio decreased due inflicting with hypercholesterolemic and hypoglycemic, but due to feeding on tubs and frint the reverse was found . Best results rented for5% , 10% groups • Serum glucose: The results of all the groups were lower at values when compared with that of control (+). Cinnamon 10% (G10) treatment indicated the more reducing in comparision all other groups including that of glimepiride drug . With total cholesterol The result showed that all of the group were of lower values when compared with control. Chicory 10% (G6) treatment gave the more reducing comparision with glimepiride drug T.G: The results showed all the groups were of lower values when campared with control chicory 10% (G6) treatment + gave the more reducing effect in camparision with other treatments including glimepiride drug HDL: the results showed that all of the groups were at higher values when campared with control group except (G3), (G4) and (G11 LDL: The results showed all the groups were of lower values when compared with control group Papaya 10% treatment gave the more reducing effect in comparision with all groups including glimepiride drug SUMMARY 127 2- Liver functions : AST: The results showed that all of the groups were at lover value when compared with control (+) group Cinnamon 10% (G10) treatment gave the more reducing comparision even in comparison with glimepiride drug ALT: The results showed that all of the groups were at lower values when compared with control group . Chicory 5% (G5) treatment gave the more reducingeffect even in comparision with gliemepiride drug Kidney Functions Urea: The results showed that all of the groups were lower of values when compared with control group. Papaya 5% (G7) treatment gave the more reducing effect even in comparision with glimepiride drug Creatinine: The results showed that all of the groups were at lower value when compared with control group (+). Cinnamon 5% (G9) caused the greatest reduction in creatinine in blood serum even in comparision with glimepiride drug Uric acid: The results showed that all of the groups were of lower value when campared with control group (+). Papaya 10% (G8) caused the highest reduction in uric acid even in compared to glimepiride drug Blood cells count The results anived at revealed that inflicting rats with< hypercholesterolemia and hyperglycemia lowered all blood cells count HB – M.C.V – M.C.H – M.C.H.C.) , while feeding on herbs and fruit SUMMARY 128 % for M.C.V., while for (HB – M.C.H. – M.C.H.C.) mixture of herbs 5%< revealed the best result Histopathological investigation : Histopathological investigation revealed that inflicting of rats with hypercholesterolemia and hyperglycemia caused distinct histological changes in liver, kidney and heart , while as in case of biological and biochemical parameters feeding of control (+) rats on test tubs and the drug ameliorated fucls changes. Best result recorded for the herbs mixture 5% and 10% where no histopathological changes were found Finally all herbs and fruit used in work were as effective as the glimepiride drug or even better.
