Abstract

In this thesis, camel pox virus vaccinal seed was firstly propagated on Vero cell line and BHK-21 cell line for 10 passages, the titer were, S.2 10glO TCIDso/ml and 4.3 log]o TCID- SO/ml respectively, so it appear that the Vero cell line was more sensitive for CPV propagation than BHK-21 cell line. Different concentrations of DEAE-dextran were used in this study, SOug/ml was completely safe on Vero cell line with high titer obtained but lower concentrations decrease the virus titer and higher concentrations gave undesirable (toxic) effect on Vero cell line. The titer of enhanced CPV with SOug Iml of DEAE- dextran after 10 passages was 6.3 log]o TCIDso/ml, while the titer of non enhanced CPV after 10 passage on Vero cell line was S.2 IOglO TCIDso/ml, this means there is 1.1 10glO increase. sixteen lab animals (8 guinea pig & 8 rabbits) were used for preparation of positive serum against CPV for using In ELISA and the titer was 1/64 when tested by ELISA for both. The result of virus titration for enhanced and non enhanced CPV also was confirmed by using ELISA on antigens prepared from enhanced and non enhanced CPV.The prepared enhanced vaccine was sterile and safe to be used in camels as documented by sterility and safety tests.
