Abstract

A total of 180 samples of salted fish 60 samples of each (salted sardine, feseikh, molouha) 30 samples of each low and high salt were examined for isolation, identification and counting of Staphylococcus aureus, isolation of salmonella, isolation of E. coli and count of total coliforms and fecal coliforms and found that all samples free from salmonella spp., E.coli and fecal coliform but during the isolation steps of Salmonella proteus, citrobacter, enterobacter, serratia, yersinia and morganella were isolated, the count of coliform in salted sardine, feseikh and molouha were1.596 loglo cfu/g, 2.6651oglO cfu/g and 2.219 loglo cfu/g respectively. 154 strains of S. aureus were isolated 47 strains from salted sardine, 55 strains from feseikh and 52 strains from molouha,All strains of S. auerus isolated were catalase positive, off positive and sensitive to novobiocine,95 strains were coagulase positive while 50 trains were coagulase negative, 115 strains were showed mannitol fermentation while 30 strains were howed mannitol non fermentation, 138 strains were DNase positive while 7 strains were DNase negative d 120 strains were showed beta heamolysis, 20 strains were showed alpha heamolysis and 5 strains
ere showed non heamolysis on blood agar.The coagulase positive S. aureus isolates were sensitive to iprofloxacin, gentamicin, streptomycin, neomycin, amikacine, vancomycine, methicillin, erythromicine, minopenicillin, tetracycline with a percentage of 100%,98%,98%,96.6%,94.9%,91.5%,84.7%,74.6% 69.5% and 59.3% respectively, and the isolates show 19 different patterns of antimicrobial resistance. 50 trains of coagulase negative S. aureus isolates show 100% sensitive to ciprofloxacin, gentamicin,
.. acin, neomycin and streptomycin, 96% sensitive vancomycin and erythromycin, 58% sensitive to ethicillin and tetracycline, and 24% sensitive to amino penicillin. when, identification of 12 isolated S.ureus (6 coagulase positive and 6 coagulase negative)were confermed by PCR by using 2 ligonucleotides primers of genes Sau234-1501 and COAG2-COAG3. Our result were shown that all olats were S. aureus all isolats were S. aureus and coagulase positive except one strain are coagulase egative. mannitole nonfermentative S. aureus isolateswhen exposure to high salt concentrations it came mannitole fermentation positive.
