Abstract
purification of 45/47kDa protein complex from culture filtrate of BCG by S1200 column , six major fractions collected , 45/47 kDa present in fraction 2, protein content was 1 mg/ml. Whole culture filtrate of M. bovis prepared. By SDS-PAGE analysis, there were band of
45/47 kDa in culture filtrates( M. bovis ,BCG) harvested after 14 days. Vaccination of
guinea pigs by 45/47 kDa, BCG, culture filtrates( M. bovis, BCG )challenged after 12 weeks.Vaccins were evaluated by humoral immune response (indirect ELISA),Cell mediated immune response (lymphocyte proliferation assay expressed by MTT). Studies of protective immunity: survival rate, root specificlung weight (RSLW).Histological appearance postchallenge by28 days. Protection against tubercles infection cannot be guaranteed by 45147kDa alone.
