Abstract

Production of live attenuated sheep pox vaccine sustained the elevated temperatures during freeze- drying, transportation storage and vaccination in unequipped tropical and subtropical zones of the world, is highly recommended, for this reason eight stabilizer formulas were individually used for preparation of eight sheep pox vaccines, which were lyophilized and then titrated and accordingly four vaccine formulas were eventually selected that should be tested for thermoprotectivity and thermostability to select the best stabilized vaccine and they were, the control vaccine that contain a stabilizer composed of (5% Lactalbumin hydrolysate :2.5% sucrose) in addition to two vaccines containing Trehalose instead of sucrose as 2.5 % and 10 % respectively and the fourth vaccine contain 10 % (wt./ V.) Trehalose alone. These vaccines were tested for sterility; vaccination and challenge test and safety in susceptible sheep. The collected blood sera were subject to serological examination for estimating the antibody response by ELISA. To test the thermal stability of the different lyophilized sheep pox Vaccine formulas they were stored at 37°C for 21days, and at 45°C for 12hours, in addition to the dissolved vaccines at 37°C for 12 hours. The results proved transcendence of sheep pox vaccines stabilized with 10% trehalose alone or in combination with 5% lactalbumin hydrolyste in the thermostability and thermoprotectivey, thereby improvement vaccination efficacy in tropical and subtropical zones which reflect positively on the farmers and national income.
