Abstract

Methotrexate therapeutic is a drug that is used to treat certain cancers Cancer cells treated with methotrexate, can not make DNA, this kills cancer cells. However, methotrexate can also be harmful to other normal cells and organs in the body, this harmful effect is called methotrexate toxicity, using methotrexate for long period and the longer methotrexate stays in the body, can increase the risk of toxicity. The Adriamycin provided therapeutic advantage when the drug was administrated intrapretoneally in patients with carcinoma, but prohibitive local toxicity limited the use of ADR via the intrapersonal route. Adriamycin damages DNA by intercalation of the anthracycline portion and inhibits topoisomerase II resulting in DNA strand breakage, generates free radicals. The cells in the S phase are the most sensitive to the cytotoxic action of ADR since it inhibits both DNA
polymerase and RNA polymerase.
