Abstract

A total number of Forty four blood samples were colleted from two different regions. All blood samples were collected from unrelated animals, moreover the sampled animals were phenotypically normal and healthy. Details of the animal sampled are as follows: 15 Zaraibi goats breed and 16 Baladi goats breed from Sakha station in Nile Delta region.
The purpose of the study was to characterize Zaraibi goats and study the genetic diversity between and within it and other two breeds of goats raised in Egypt by using eight microsatellite markers. These breeds are: Baladi and Sinawi. The relationship and the purity of these breeds will be also evaluated.
In the present study eight microsatellites were selected from the available list of the microsatellites suggested by International Society of Animal Genetics (ISAG) for estimation of genetic diversity in Caprine. The origins of these microsatellites were from Caprine: SR-CRSP5, SR-CRSP8 whiles the microsatellites: INRA063, ILSTS87, INRA023, ILSTS19 were from Bovine origin, finally the microsatellites OarFCB20, McM527 were from Ovine origin.
The DNA were isolated and purified from the blood samples using salting out procedure. The polymerase chain reactions (PCR) were performed using specific primers for the microsatellites studied. The PCR products were characterized on polyacrylamide vertical electrophoresis under denaturing conditions, stained with fluorescent ethidium bromide.
Photos were captured using gel documentation system and analyzed with specific software. The results were statistically analyzed using POPGENE and FSTAT software.
