Abstract

The aim of this study was to investigate the impact of HPV high-risk genotypes DNA integration on HNSCC
development in Egyptian patients through:
A) Direct detection of the HPV DNA in the genome of cancer tissues and comparing it with safety margin tissues from the same patient to prove that the insult of integration of the virus in the genomic DNA is linked to the process of malignant transformation as a causeeffect relationship.
B) Studying the expression of the cIAP-2in a trial to link HPV integration with the anti-apoptotic cIAP-2 gene over-expression.
C) Investigating a possible synergistic role of the viral genome integration and other environmental factors such as smoking.
This study included 30 patients who were examined endoscopically and the tumor was resected by hot loop
resection and sent for pathological staging and grading of the tumor according to (TNM) classification. Their tissue samples were stratified into two groups:
Group A: Head and neck cancer tissues.
Group B: safety margin of the cancer tissues of the same patients as our control samples.
All patients in our study were subjected to complete detailed history taking, general and local examination, and routine laboratory investigations. All patients were subjected to qualitative detection of HPV gene in their tissue samples by viral DNA extraction and detection using PCR technique.
