Abstract

This work was carried out to study trihexphenidyl effects on albino rats and evaluate clinical presentations of animals during intake of drug and try to estimate LD50 of Parkinol (trihexphenidyl) and estimate LD50 in dependent animals The study was carried out on 60 adult male albino rats of an average weight (150 - 200 gm) obtained from a local animal breeding house Rats were divided into 3 main groups as follow Group I: Control group consisted of 10 rats. This group did not receive any treatment Group II: non dependent LD50 group (10) rats were given LD50 of trihexphenidyl, it equals = about 50-54 mg (250-270mg/kg) in rat weighting 200 gm Group III Dependent group consisted of 40 rats that were given trihexphenidyl in gradually increasing doses until they reached the dependence dose in one month then divided into 4 subgroups Subgroups III a: Ten rats of the dependent group. (Control of the dependent group Subgroups III b: Ten rats were given LD50 of trihexphenidyl Subgroup III c: Ten rats were given 100 mg of trihexphenidyl (double LD50 Subgroup III d: Ten rats were given 150 mg of trihexphenidyl (triple LD50 All groups were closely observed over one month for general condition and behavior At the end of experiment control group, dependent group and lived rats of LD50 were sacrificed after cervical dislocation and autopsy was done to obtain the internal organs, liver and brain for histopahtological study Blood samples were also taken for biochemical study as liver and kidney function tests and complete blood count.
