Abstract

The present study aimed to investigate the chronic toxicity of profenofos and fenarimol on rats and determined the potential action of both pesticides on general health. These study was conducted on 140 mature male albino rats divided into seven groups group (I) served as control group which received corn oil only, group (II) and (III) given profenofos 1/10 LD50 and 1/20 LD50 respectively, group (IV) and (V) given fenarimol 1/10 LD50 and 1/20 LD50 respectively and group (VI) and (VII) given 1/10 LD50 profenofos plus 1/20 LD50 fenarimol and 1/10 LD50 fenarimol plus 1/20 LD50 profenofos respectively .All groups treated orally one dose every week for six month. Whole blood, serum, bone marrow, testis, liver and kidney tissues were collected after 1st, 2nd, 3rd and 6th month. Whole blood collected for hematological investigation (RBCs, WBCs, Hb concentration, PCV%). Serum was collected for biochemical analysis. Bone marrow was collected for detection of chromosomal aberration . Testis and tail of epididmes collected for spermogram. Samples of liver and kidneys were collected for residue analysis, and tissue samples of liver, spleen, kidney, brain, testis, heart and lung were collected for histopathological examination. The results revealed a significant decreased in mean body weight of rats received profenofos and\or fenarimol . These reduction was clear in groups treated by high doses .The relative organs weight of liver and kidneys of treated groups showed a significant increase, while relative spleen weight showed a significant decrease (P<0.05)…RBCs, WBCs, Hb concentration and PCV% of treated rats were highly significantly decrease (P<0.01) . Total serum protein and albumin levels of treated rats were significantly decreased (P< 0.05) .The significances were prominent after the 6th month of treatment specially in groups received mixed pesticides treatment. The study recorded a highly significant elevation (P<0.01) of AST , ALT ,Alkaline phosphatase, GGT ,Urea and Creatnine in the sera of all treated rats specially at the last month of treatments received high dose and mixed two pesticides. While the AchE activity as a biomarker of organophosphrous insecticide showed a highly significant decrease (P<0.01) in group of high dose of profenofos and groups received the two pesticides, The serum Alfa-fetoprotein level showed highly significant increase (p<0.05) especially in groups of high dose and groups received the two pesticides after the 6th month of treatment. Regarding to the effect of profenofos and fenarimol on sperm cell count and morphological abnormalities there were a significant decreased (P<0.05) of sperm cell count and elevation of sperm abnormalities specially of groups treated by two pesticides and after the 6th month of treatment . Concerning to genotoxic effect of profenofos and fenarimol a highly significant elevation (P<0.01) of micronuclus % specially groups received high dose and groups treated by two pesticides were detected>Chromosomal aberrations in bone marrow showed a significant increase in structural chromosomal aberrations (deletion breach, fragment, acentromer, rings and gaps). Also a high significant increase
