Abstract

The present project was planned to synthesize some new pyridine derivatives as well as their condensed heterocyclic derivatives andto study their reactions with a variety of reagents hoping to get novel compounds with anticipated biological activities. However the work included in this thesis can be divided into two main parts as described below.
The first part includes synthesis and of new fluorine-containing pyridins furopyridines and thienopyridines as well as their condensed heterocyclic derivatives.
Our approach to synthesis of the target compounds started from 3-cyano-6-(2-thienyl)-4-trifluoromethylpyridine-2(1h)-one (508) and its thiono analogue 56 which were prepared by cyclocondensation reaction of (2-thenoyl)trifluoro-acetone (55) with cyanoacatamide or cyanoacatamide
When compound 508 eas reacted with ethyl chloroacetate, in dry acetone containing anhyd. K2co3 and nal, the pyridine derivative 509 was obtained. Cyclization of 509 to furo (2,3-b)- and Acoh-h2so4 mixture similarlt, the compounds 616and 617 were prepared
In the second part of the thesis, synthesis and reactions of some new pyridothienopyrimidines, pyridothienopyrimidobenz- imidazole and condensed phrazolopyridines are reported.
5-Acetyl-3-cyano-4-(4-methoxphenyl)-6-methylpyridine-2(1h)thione (94)was used as starting compound in this part. Thus, its reaction with methyl iodide chloroacetamide or-2-chloromethy-1h-benzimidazole, in the pressncce of sodium acetate gave pyridine derivative 623, 618 or 625 respectively it underwent intermolecular Thorpe- Ziegler cyclization to giveb thieno(2,3-b)pyridine derivative 619 or 626 respectively.
