Abstract

Since its identification in the early 1990s, peroxisome-proliferator-activated receptor γ (PPAR-γ), a nuclear hormone receptor, has attracted tremendous scientific and clinical interest. The role of PPAR[gamma] in macronutrient metabolism has received particular attention, for three main reasons: first, it is the target of the thiazolidinediones (TZDs), a novel class of insulin sensitizers widely used to treat type 2 diabetes; second, it plays a central role in adipogenesis; and third, it appears to be primarily involved in regulating lipid metabolism with predominantly secondary effects on carbohydrate metabolism, a notion in keeping with the currently in vogue ‘lipocentric’ view of diabetes. So this work was designed to try to understand on molecular level the role of PPAR- γ in glucose homeostasis and study the effect of PPAR- γ agonist on insulin receptor substrate and insulin levels also the effect of PPAR-γ agonists on the mechanism of action of insulin.A total of 60 Male Sprague-Dawley rats, 150 + 10 g B.W. were used in this study, Rats were accommodated at the animal house in faculty of vet. Med. Zagazig university for 15 days before the experiment, and were kept on standard diet, and they randomly divided in 5 groups, 1-Control group (non-treated group) Fed standard diet and maintained for 2 months.
2-Streptozotocin treated group, Received single dose of streptozotocin at the beginning of the experiment by interperitoneal injection of 65 mg/kg body wt dissolved in citrate buffer (0.01 M, pH4.5), rats were supplied with 5% glucose solution in their drinking water, as it is known that hypoglycemia occurs 7-8 hours after STZ injection. Then kept on standard diet for the rest of two months (Bolkent et al, 2000). 3-Streptozotocin and Insulin treated group, Fed standard diet received single dose of streptozotocin at the beginning of the experiment by intraperitoneal injection of 65 mg/kg body wt dissolved in citrate buffer
