Abstract
This work was carried out in the Plant Tissue Culture Laboratory in Fac. Envilon. Agric. Sci., El-Arish, Suez Canal Univ. during the period from 2002 to 2005. The aim objectives of these studies were to establish callus formation for Achillea fragrantissima and Ecballium elaterium by using tissue culture technique and to study their response to salinity and drought stresses and the effect of these stresses on callus mineral composition. The results show that the best callus fresh weight and callus index were obtained with 2 mgl1 2,4-D for the two plant species. Increasing NaC1 levels and mannitol concentrations decreased callus fresh and dry weight. Adding 4 mgF’ BA to MS medium in E. elateriwn gave the highest No. shoots and No. roots, but, A. fragrantissima did not response. The combination between Full MS 1.5 mgF BA and 0.2 mgi’ NAA gave higher number and length of shoots in A. fragrantissima.
