Abstract

To date the nasal insulin administration is considered to be the most promising route among all non-parenteral routes of insulin delivery due to, its convenience, tolerability and its rapid rate of absorption mimikingmore closely the jnsulin-response to meal in normal subjects. The aim of the present work was to study the effect of intranasal lispro insulin versus crystalline human insulin using azone as a potent penetration enhancer on plasma glucose and serum insulin levels of type 2 diabetic patients. The work was carried out on 20 adult male type 2 diabetic patients. . Each patient acted as his, own control. Azone (1¬dodecylazacyc1oheptan-2-one) (Upjohn Company) used as 2% solution in propylene glycol. Insulins used were HumulinR insulin 1 OOU/ml. ”RNA recombinant biosynthetic” and lispro insulin 100U/ml (Elil.illy and Co., Indiana polis, USA). They were sprayed using a special precaliberated : nasal spraying device .. Each puff delivered 0.1 ml (10 units insulin). All patients were hospitalized at the Diabetes and Metabolism Unit, Alexandria Main University Hospital. Patients with hepatic, renal or cardiac diseases were excluded. Drugs known to affect glucose homeostasis or have local effects on the nasal mucosa were also excluded.
