Abstract
Concept of this work is the preparation and purification of meningococcal capsular polysaccharide vaccine (A&C) and its formulation to produce a locally bivalent meningococcal capsular polysaccharide (A+C) vaccine in VACSERA.
The MCPS A+C prepared from Neisseria meningitidis strain A1 and C11 which were obtained from Dr. Carl Frash, Office of Drugs and Biologics, FDA, Bethesda, MD, USA. Strain A1 and C11 were grown on Mueller-Hinton plates and selected colonies are inoculated on modified Frantz media. The bacterial cells separated from the culture by centrifugation and the clear supernatant was purified by different purification procedure to obtain a highly purified non toxic and non pyrogenic CPS.
The protein content and nucleic acid found to be with the range of WHO requirements. The MCPSA and MCPSC were fractionated on sepharose CL-4B and sepharose 4B columns for determination of Kd which also, found with the range < 0.4.
